The new 27 Jane South bus route will operate between Jane Station on Line 2 Bloor–Danforth and Mount Dennis Station on Line 5 Eglinton via Jane Street.

Northbound: From Jane Station bus terminal via south on Armadale Avenue, west on Bloor Street West, north on Jane Street, east on Eglinton Avenue West, south then west on Keelesdale Road, and north on Photography Drive to Mount Dennis Station bus terminal.
Southbound: From Mount Dennis Station bus terminal via south on Photography Drive, east then north on Keelesdale Road, west on Eglinton Avenue West, south on Jane Street, and east into Jane Station bus terminal.
