The 51 Leslie bus route operates between Leslie Station on Line 4 Sheppard and Donlands Station on Line 2 Bloor–Danforth, with a connection to Line 5 Eglinton at Laird Station.

Northbound 51A: From Donlands Station bus terminal, east on Strathmore Boulevard, north on Donlands Avenue, north on Millwood Road, north on Laird Drive, east on Eglinton Avenue East, north on Leslie Street, east and north through the North York General Hospital grounds, west and south via the Leslie Street overpass, west on Esther Shiner Boulevard, and north on Old Leslie Street to Leslie Station bus terminal.
Southbound 51A: From Leslie Station bus terminal, south on Old Leslie Street, east on Esther Shiner Boulevard, north and east via the Leslie Street overpass, south and west through the North York General Hospital grounds, south on Leslie Street, west on Eglinton Avenue East, south on Laird Drive, south on Millwood Road, and south on Donlands Avenue to Donlands Station bus terminal.
51B Leslie (Except Sunday Early Morning)
Northbound 51B: From Donlands Station bus terminal, east on Strathmore Boulevard, north on Donlands Avenue, north on Millwood Road, north on Laird Drive, east on Eglinton Avenue East, north on Leslie Street, east on Lawrence Avenue East, north and east on The Donway West, and east and south on The Donway East to The Donway East at Lawrence Avenue East.
Southbound 51B: From The Donway East at Lawrence Avenue East, south and west on The Donway East, west and north on The Donway West, west on Lawrence Avenue East, south on Leslie Street, west on Eglinton Avenue East, south on Laird Drive, south on Millwood Road, and south on Donlands Avenue to Donlands Station bus terminal.

Northbound 51B (Sunday Early Morning): Out of service from Greenwood Station bus terminal, east on Strathmore Boulevard, south on Linnsmore Crescent, west on Danforth Avenue, entering service via north on Donlands Avenue, north on Millwood Road, north on Laird Drive, east on Eglinton Avenue East, north on Leslie Street, east on Lawrence Avenue East, north and east on The Donway West, and east and south on The Donway East to The Donway East at Lawrence Avenue East.
Southbound 51B (Sunday Early Morning): From The Donway East at Lawrence Avenue East, south and west on The Donway East, west and north on The Donway West, west on Lawrence Avenue East, south on Leslie Street, west on Eglinton Avenue East, south on Laird Drive, south on Millwood Road, south on Donlands Avenue, east on Danforth Avenue to Byron Avenue, then out of service east on Danforth Avenue and north on Linnsmore Crescent to Greenwood Station bus terminal.
